Pre-Lab Questions
1. On which plate(s) would you expect to find bacteria most like the E. coli on the source plate? Explain. The plates you would expect to find bacteria most like the E. coli on the source plate are the -DNA plates which is also the control plates because they have not been altered with DNA.
2. On which plate(s) would you find only genetically transformed bacterial cells? Why? The plates you find only genetically transformation bacterial cells would be the +DNA +AMP +IPTG because the IPTG acts like a inducible promoter and helps regulate the GFP expression.
3. What is the purpose of the control plates? Explain the difference between the controls and why each one is necessary. The growth of the -DNA should be normal because it has unaltered E. coli on agar which is also unaltered. The -DNA +AMP plate has both the unaltered E. coli and ampicillin, therefore we are shown how resistant the E. coli is present with the ampicillin around.
